Stres Oksidatif dan Antioksidan

Kita sering mendengar tentang stres yang bisa memicu berbagai macam penyakit. Stres berperan dalam banyak hal yang tentunya merugikan bagi tubuh kita. Selain stres, dalam dunia medis khususnya farmasi kita sangat sering mendengar senyawa dengan kandungan antioksidan. Bahkan didalam iklan yang sering ditayangkan di televisi juga sering terdengar promosi yang menyatakan bahwa obat-obat herbal tersebut memiliki kandungan antioksidan yang baik bagi tubuh. Pertama-tama kita akan membahas mengenai stres oskidatif. Apa itu stres oksidatif ?         Menurut wikipedia, stres oksidatif adalah keadaan di mana jumlah radikal bebas di dalam tubuh melebihi kapasitas tubuh untuk menetralkannya . Akibatnya intensitas proses oksidasi sel-sel tubuh normal menjadi semakin tinggi dan menimbulkan kerusakan yang lebih banyak. Radikal bebas adalah molekul yang kehilangan satu buah elektron dari pasangan elektron bebasnya, atau merupakan hasil pemisahan homolitik suatu ikatan koval...

Oxamniquine terhadap Schistosomiasis


Schistosomiasis adalah infeksi dengan cacing darah dari genus Schistosoma, yang diperoleh transcutaneously dengan berenang atau berendam di air tawar terkontaminasi. Organisme yang menginfeksi pembuluh darah dari GI atau sistem GU. Gejala akut adalah dermatitis, diikuti beberapa minggu kemudian oleh demam, menggigil, mual, sakit perut, diare, malaise, dan mialgia. Gejala kronis bervariasi dengan spesies tetapi meliputi diare berdarah (misalnya, dengan mansoni S.) atau hematuria (misalnya, dengan haematobium S.).

Obat ini adalah antihelmintik yang memiliki aktivitas melawan Schistosoma mansoni. Oxamniquin agen dosis tunggal yang ampuh untuk pengobatan infeksi S. mansoni, dan menyebabkan cacing bergeser dari pembuluh darah ke hati, dimana cacing jantan tetap tertahan dihati dan cacing betina kembali ke mesentrium, tapi tidak bisa lagi melepaskan telur.

Mekanisme Kerja

    menurut saya belum tentu karena oxamniquine efektif terhadap S.mansoni tetapi belum tentu efektif terhadap jenis cacing lain

    1. Saya setuju dengan pendapat kak fen. Oxamniquine terus digunakan dalam program pengendalian S. mansoni, terutama di Amerika Selatan. Sedangkan Obat lini kedua praziquantel untuk pengobatan schistosomias .

    3. menurut saya kak, oxamniquine bukan dijadikan pilihan pertama terhadap pengobatan schistosomiasis namun lebih ke terapi dengan prazikuantel. hal ini dikarenakan prazikuantel efektif pada semua infeksi cacing daun (trematoda) kecuali Fasciola hepatica yang menyebabkan skistosomiasis

    4. saya setuju dengan pendapat Dyah, Pengobatan spesifik biasanya digunakan Praziquantel (Biltricide®) karena obat adalah pilihan untuk semua spesies. sedangkan Oxamniquine merupakan obat alternatif untuk S. mansoni.

  2. saya akan menjawan no 2 Oxamniquine sekarang diketahui menghambat sintesis asam nukleat di sel skistosomal. Mekanisme aksinya diduga melibatkan aktivasi enzim sulphotransferase yang ada pada sel parasit.

    Saya akan mmjawab prtnyaan no 7 menurut artikel yg prnh saya baca Saat ini ada dua obat yang tersedia, Praziquantel dan Oxamniquine , untuk pengobatan schistosomiasis. Karena biaya yang lebih rendah per perawatan, Praziquantel umumnya dianggap sebagai pilihan pertama untuk pengobatan.

    1. Dikhawatirkan obat ini akan menembus plasenta janin. Selain itu ditakutkan obat ini dapat mengganggu pertumbuhan dan perkembangan janin yang dapat menimbulkan resiko bayi lahir cacat atau prematur

    menurut saya bagian penting pada oxamniquine terdapat pada rantai alkilnya. rantai alkil meningkatkan aktivitas. Penjelasannya adalah bahwa percabangan meningkatkan interaksi van der Waals ke daerah hidrofobik. Sebagai alternatif, meningkatkan lipophilicity obat, sehingga memudahkan perjalanan melalui membran sel.
